How I do it:
During the week I search Ebay Pulse to see what is the hottest things selling at the moment. This is free, and it will tell you what people are searching for.
Saturday Mornings is spent out at Yardsales, or thrift stores getting things to sell. Once I get the items that I will have to sell for the week, I spend the next few hours taking pictures and writing descriptions about it. The better the photos, and the more descriptive you can make the item, the more money you will get for that item.
Once I have the photos and the descriptions I am ready to post. To make really professional looking Ebay Listings I use the free site Auctiva. If you are wondering why, I think this photo from their site says it all.

The thing that you want to know about ebayer's is that they expect their packages shipped the next day after they pay for the item. If you don't want to go to the post office daily, then you simply put in your listing, shipping will only be done on this day and this day. Now you won't have to go to the post office daily and you won't get emails from people asking where their packages are.
